چکیده
Placenta! tissues were obtained from Chinese women in Tai wan who had been exposed to contaminated rice oils containing polychlorinated biphenyls and their thermal degradative prod ucts. Exposure via the diet occurred 4-5 years prior to preg nancy. Placental microsomal fractions from eight of the nine exposed subjects studied showed marked elevation of benzo(a)pyrene hydroxylation and 7-ethoxyresorufin 0-deethylation activities relative to control subjects. Placental microsomes from exposed subjects were found to contain a protein that cross-reacted with antibodies raised to rabbit cytochrome P-450 isozyme 6, an isozyme induced by polycyclic aromatic hydrocar bons. This protein was not observed with microsomal samples from control subjects. A significant correlation was found be tween the relative amounts of the immunoreactive protein and benzo(a)-pyrene hydroxylation and 7-ethoxyresorufin 0-deethylation activities. The 7-ethoxyresorufin 0-deethylation activities were inhibited by a-naphthoflavone, a compound known to inhibit activities of rabbit cytochrome P-450, isozyme 6.

Placental markers of human exposure to polychlorinated biphenyls and polychlorinated dibenzofurans.
Our studies have evaluated biochemical changes in placentae from humans exposed to rice oil contaminated with polychlorinated biphenyls (PCBs) and polychlorinated dibenzofurans (PCDFs) in Taiwan. Placentae were obtained from nonsmoking women 4 to 5 years after the exposure had occurred. The exposed individuals ingested approximately 1 to 3 g PCBs and 5 mg PCDFs, and many exhibited symptoms char...

Individual blood samples obtained from yusho and yu-cheng patients who had been poisoned by ingesting contaminated cooking oils, from workers occupationally exposed to polychlorinated biphenyls (PCBs) and from unexposed individuals were analyzed for PCBs, polychlorinated quaterphenyls (PCQs) and polychlorinated dibenzofurans (PCDFs) by gas chromatography and mass spectrometry. PCBs were found i...
متن کاملPolychlorinated dibenzofurans--potent inducers of rat hepatic drug-metabolizing enzymes.
The effects of polychlorinated dibenzofurans (PCDFs), trace toxic contaminants of commercial polychlorinated biphenyl preparations (PCBs), on the induction of hepatic drug-metabolizing enzymes were studied in the rat. PCDFs were about a thousand times more potent than PCBs (Kanechlor-500) as inducers of cytochrome P-450. Rats given 10 microgram/kg of PCDFs intraperitoneally for 3 days showed si...
متن کاملParticipation of cytochrome P-450 in reductive metabolism of 1-nitropyrene by rat liver microsomes.
Reductive metabolism of carcinogenic 1-nitropyrene by rat liver microsomes and reconstituted cytochrome P-450 systems was investigated. Under the nitrogen atmosphere, 1-aminopyrene was the only detected metabolite of 1-nitropyrene. The reductase activity in liver 105,000 X g supernatant fraction was ascribed to DT-diaphorase, aldehyde oxidase, and other unknown enzyme(s) from the results of cof...
متن کاملHydroxylation of prostaglandins by inducible isozymes of rabbit liver microsomal cytochrome P-450. Participation of cytochrome b5.
The hydroxylation of prostaglandin (PG) E1, PGE2, and PGA1 was investigated in a reconstituted rabbit liver microsomal enzyme system containing phenobarbital-inducible isozyme 2 or 5,6-benzoflavone-inducible isoenzyme 4 of P-450, NADPH-cytochrome P-450 reductase, phosphatidylcholine, and NADPH. Significant metabolism of prostaglandins by isozyme 2 occurred only in the presence of cytochrome b5....
